No not needed. You can continue your (current) career.

For what I tested the mods will be applied immediately after you click on next-day button. IE if I have energy regen setting on start, then change values later, will I see those values/results in my game as I progress or is it locked to the initial save values on career generation/first save?
Sorry to ask a schoolboy question but I just never fully understood that part. Im alwyas reluctant to start a serious full manager career due to updates, but Im wondering if im waiting for nothing.

I am playing as arsenal, I offered Diego from wolfsburg 140k wages when his base wage is 80k and he says no? Also offered Piszczek from dortmund 100k wages when his base is 60k and he also said no!!! Same happened with chiellini for juventus.

Also, have you made it easier to get other jobs? You normally just get rejected by everyone.
